It is the early 1880s and Dr Frankenstein’s evil granddaughter Maria (Narda Onyx) has moved to the American West with her brother Rudolph (Steven Geray) and is utilising the prairie lightning storms in her experiments on immigrant children snatched from a dying town.
While Maria is happily killing the children and replacing their brains with artificial ones, (intending to revive them as her slaves), Rudolph is reluctant to help his sister but is too afraid of her to do otherwise.
Meanwhile, Mañuel Lopez (Felipe Turich) and his wife Nina (Rose Turich) are leaving town with their daughter Juanita (Estelita Rodriguez) because of the frequent disappearances – the latest of which was their son.
Gunslingers Jesse James (John Lupton) and Hank Tracy (Cal Bolder) arrive in town and hook up with Butch Curry (Roger Creed), the head of a local gang called The Wild Bunch, with the intention of stealing $100,000 from the next stagecoach.
Butch’s own brother Lonny (Rayford Barnes), goes to the Marshal (Jim Davis) and reveals the plot in exchange for becoming his deputy and claiming the reward for the capture of Jesse James.
During the robbery, the Marshal and his men shoot the two remaining members of the Wild Bunch and seriously wound Hank.
Jesse and Hank escape and stop at the Lopez’s campout to tend to Hank’s wound and sleep until the morning. During the middle of the night, Juanita wakes up Jesse and Hank and leads them back to town to the Frankensteins’ house to fix up Hank despite her parents forbidding her to go there.
Maria agrees to help, and even covers for her guests when the Marshal and Lonny come around looking for them – but her actual plan is to use Hank for another of her experiments, transforming him into her servant, now called Igor.
Shot in just eight days, Jesse James Meets Frankenstein’s Daughter was produced as part of a double feature with Billy the Kid Versus Dracula (1966) which was also directed by William Beaudine.
